I have a visitor visa b2 from India. My brother is sponsoring for me. Will my b2 visa be canceled when he does?

Not necessarily but be sure to disclose that an immigrant visa petition has been filed on your behalf if you are ever asked such as on a new visa application or at the Port of Entry. You will have a slightly higher burden to prove that you intend to go back to India and honor the terms of the B-2 (such as job to return to children or spouse, important upcoming event, etc.)
