I was petitioned by my mother and was approved 2004. I applied for a tourist visa and was denied 2 years ago. Will this affect my petition?

It won't as long as there was no fraud charge.

Whether a denial of a visa application will affect you ability to immigrant to the U.S. based on your mother's petition depends on why the visa was denied. If it was denied because you submitted false information (ex. your marital status, whether you had relatives in the U.S., the reason for your trip, etc.), they it could affect you. If it was denied because the consular officer did not believe you would return, then it should not affect your petition.
