I am almost certain that this music is unique. How can I protect the song while looking to sell to one of these advertising agencies?

You must register it federally as a copyrighted work. It costs about $40. You can do it on line. While all works of art are automatically copyright protected, if you don't federally register it within 90 days you lose some very important rights with regard to what remedies you can sue for.
